In the last posting, we shared about how to sterilize Arabidopsis seeds. This time we will share about how to sterilize rice seeds. Probably, this method is also applicable for other cereals grain such as maize, barley, wheat even soy bean. You can modify this method little bit for your convenient. The whole steps are:
Put 10-20 rice seeds into 2 mL E-tube.
(option) Add 1 mL Ethanol to tube, mix with low vortex for 1 minute, then discard the Ethanol.
Add washing solution (bleach) around 3/4 of tube, or you can add about 1 mL. Note: Bleach solution is made from a half concentration of Clorox with sterilized water. (option) You can add 0.01% Tween 20 into it.
- Mix well using vortex with low strength for 30 minutes up to 1 hour.
- Pipet up the washing solution and change with sterilized water, mix, and change again with the same water. Repeat this steps for 5 or 6 times.
- Add 1 mL sterilized water and store the seeds in 4 degree celcius for overnight, or
- You can remove all water from seed, put in the sterilized tissue towel, and directly put into media (seed plating).
